In rats, ingested phloretin is converted to phloretin by hydrolytic enzymes in the small intestine.Phloretin hydrolase hydrolyzes phloretin into phloretin and phloroglucinol.

In animal models, phloretin inhibits the active transport of glucose into cells through SGLT1 and SGLT2,although this inhibition is weaker than that of its glycoside phloridin.One of its important roles is to inhibit the absorption of glucose by the small intestine and the reabsorption of glucose by the kidney.Phloretin also inhibits multiple urea transporters.When combined with a high-protein diet, it causes urea loss and diuresis.Phloretin has been found to suppress body weight gain and improve metabolic homeostasis in high-fat diet-fed mice.Phloretin inhibits aquaporin 9 (AQP9) on mouse hepatocytes.

Phloretin-functionalized gold nanoparticles (Pht-GNPs) were synthesized using a single-step synthesis method and tested for their anticancer activity.Pht-GNPs showed significant cancer cell toxicity compared with free phloretin.
